auto slime in bike inner tube tire

Whenever you're dealing with a slime-filled tube and have to do anything with the valve (including air the tire up, step one is always to leave the tire with valve stem at the top, around the 1-o'clock position, until the slime runs down to the bottom of the tube.

How long that takes depends on the temperature, but not long even if it's a little chilly.

On my old gallon jug of it I used to have, that was part of the instructions printed on it.
 
if you use slime stick 2 or 3 small cable ties around the valve stem, worked this out when my stem blew out at a garage while pumping up the tyres, other alternative was a 3-4 kay push all uphill.
that was the last time i used it, now its old racer tyres trimmed to fit as tyre liners, and old thorn resistant tubes slit to wrap around the new tube, 2 layers of protection, no punctures of slow leaks since.
 
I find the car stuff works loads better, I know the stuff that comes already in tubes is complete crap as mates have gotten punctures with it before, but the car stuff put into tubes works wonders.
